    Notes: Abstract The processes of transport differentiation from stem cell to the terminally differentiated cell in intact colonic crypts are difficult to study because access to the lumen is limited. Colonocytes were isolated from the lower two-thirds of rat distal colon crypts and grown to confluence on reconstituted basement membranes and permeable support in primary culture. Crypt and surface cells were distinguished by the uptake of [3H]thymidine and [3H]leucine and by brushborder fluorescence binding. Ion concentrations in apical and basolateral compartments of filter monolayer cultures after 48 h of incubation on days 16–18 were (in mM): apical, Na+ 116±4 (n=48) and K+ 6±1 (n=48); basolateral, Na+ 151±3 and K+ 3.7±0.5, respectively (mean ±SE). Aldosterone (10−8 M), added to the basolateral compartment from days 10–18, changed apical Na+ to 72±6 mM and apical K+ to 13±4 mM (n=23). Dexamethasone (10−8 M) changed apical Na+ to 84±7 mM but did not influence apical K+ (n=22). Transmonolayer electrical potential difference (V tM; control medium; days 8–10) was 5±1 mV (n=16; apical compartment negative); electrical resistance (R tM) was 217±21 Ω · cm2 and short circuit current (I SC) was 21±5 μA · cm−2. Amiloride (0.1 mM; n=12) in the apical medium decreased V tM to 2±1 mV and I SC to 11±4 μA · cm−2. Aldosterone (10−8 M) after 1 week in the basolateral compartment (n=21) changed V tM to 12.3±3 mV, R tM to 92±9 Ω · cm2, and I SC to 138±23 μA · cm−2. Apical amiloride (0.1 mM; n=9) decreased the induced V tM to −3±1 mV and I SC to −13±7 μA · cm−2. Colonic-crypt-derived epithelial cells proliferate and differentiate in primary culture, when grown on reconstituted basement membrane substratum and in supplemented medium, to form monolayers that express net Na+ absorption and net K+ secretion after 1 week. Na+ and K+ vectorial transport differentiation is primarily regulated by aldosterone, which specifically induces apical conductive Na+ transfer. Mineralocorticoid and glucocorticoid hormones appear to have differing actions on ion transport in functionally surface-type colonocytes derived in culture from isolated crypt-type cells.

    Notes: Abstract A transcription unit petCB from Chlorobium limicola is described. The leading gene petC codes for a Rieske FeS-protein of 19.04 kDa with 181 amino acid residues. The following gene petB codes for a cytochrome b of 47.48 kDa with 428 amino acid residues. The transcription unit lacks a third gene pet-A for cytochrome c 1 or-f, which is found in the fbc-operons of gram-negative bacteria. In the derived amino acid sequence for the Rieske FeS-protein the four cysteines and the 2 histidines are conserved in the peptides binding the 2Fe2S-cluster, although the redox potential of the cluster is about 150 mV more negative in Chlorobium. The gene for cytochrome b includes the coding region for an N-terminal, positively charged extension which is typical for Chlorobium. The gene is not split into two parts for cytochrome b 6 and subunit IV. However, a fourteenth amino acid between the two histidines in the fourth, putative transmembrane helix, and the lack of an eighth transmembrane helix at the C-terminus, among other features, clearly resemble the cytochrome b 6 f-complexes. Therefore, the separation into b 6 f- and bc 1-type complexes during evolution must have occurred before the split of the gene.

    Description: Die im Rahmen dieses Beitrages zur Vorbereitung, Planung und Realisierung von Unternehmensnetzwerken betrachteten Aspekte, Ursachen, Bedingungen und Erfahrungen aus der Evaluierung eines extrem komplizierten, strategisch angelegten Strukturierungsprozesses im Bereich der kleinen und mittleren Unternehmen (KMU) fundieren primär auf drei Quellen. Erste Quelle ist eine Studie des Institutes für Strukturpolitik und Wirtschaftsförderung Halle-Leipzig (isw) zu "Netzwerken in Sachsen-Anhalt"[1], die Quellen [2] und [3] sind Forschungsberichte zu den Themen "Aufbau eines Netzwerkes zur Verwirklichung des ökologisch-ökonomischen Bauens unter Beteiligung von kleinen und mittleren Unternehmen verschiedener Branchen" in der Region Halle-Dessau-Bitterfeld-Wolfen [2] und "Aufbau eines absatzorientierten Netzwerkes kleiner und mittlerer Unternehmen im Raum Leuna-Merseburg"- auch als Chemiedreieck bezeichnet [3]. Die beiden Netzwerkprojekte wurden vom isw initiiert und planungsseitig vorbereitet, werden vom isw wissenschaftlich begleitet und vom Land Sachsen-Anhalt gefördert.

    Description: Background: Acholeplasma oculi belongs to the Acholeplasmataceae family, comprising the genera Acholeplasma and 'Candidatus Phytoplasma'. Acholeplasmas are ubiquitous saprophytic bacteria. Several isolates are derived from plants or animals, whereas phytoplasmas are characterised as intracellular parasitic pathogens of plant phloem and depend on insect vectors for their spread. The complete genome sequences for eight strains of this family have been resolved so far, all of which were determined depending on clone-based sequencing. Results: The A. oculi strain 19L chromosome was sequenced using two independent approaches. The first approach comprised sequencing by synthesis (Illumina) in combination with Sanger sequencing, while single molecule real time sequencing (PacBio) was used in the second. The genome was determined to be 1,587,120 bp in size. Sequencing by synthesis resulted in six large genome fragments, while the single molecule real time sequencing approach yielded one circular chromosome sequence. High-quality sequences were obtained by both strategies differing in six positions, which are interpreted as reliable variations present in the culture population. Our genome analysis revealed 1,471 protein-coding genes and highlighted the absence of the F1FO-type Na+ ATPase system and GroEL/ES chaperone. Comparison of the four available Acholeplasma sequences revealed a core-genome encoding 703 proteins and a pan-genome of 2,867 proteins. Conclusions: The application of two state-of-the-art sequencing technologies highlights the potential of single molecule real time sequencing for complete genome determination. Comparative genome analyses revealed that the process of losing particular basic genetic features during genome reduction occurs in both genera, as indicated for several phytoplasma strains and at least A. oculi. The loss of the F1FO-type Na+ ATPase system may separate Acholeplasmataceae from other Mollicutes, while the loss of those genes encoding the chaperone GroEL/ES is not a rare exception in this bacterial class.
