    Notes: Abstract Cowpea protoplasts were prepared from plants of different ages and examined for their ability to take up polyamines and for their infectibility by alfalfa mosaic virus. A lag period of ∼20 h was necessary before the onset of rapid polyamine uptake; the occurrence of this rapid uptake depended on the age of the leaves used for protoplast preparation. The percentage of infection of cowpea protoplasts by alfalfa mosaic virus, and the amount of virus produced also depended on the age of the plants used for protoplast preparation. In contrast, the uptake of amino acids was rapid in all cowpea protoplasts tested.

    Notes: Abstract Ribosomal frameshifting on retroviral RNAs has been proposed to be mediated by slippage of two adjacent tRNAs into the — 1 direction at a specific heptanucleotide sequence. Here we report a computer-aided analysis of the structure around the established or putative frameshift sites in a number of retroviral, coronaviral, toroviral, and luteoviral RNAs and two dsRNA yeast viruses. In almost all cases a stable hairpin was predicted four to nine nucleotides downstream of the shifty heptanucleotide. More than half of the resulting hairpin loops give rise to potential pseudoknotting with sequences downstream of this hairpin. Especially in the case of the shifty heptanucleotides U UUA AAC and G GGA AAC, stable downstream pseudoknots are present. Indications were also found for the presence of pseudoknots downstream of amber stop condons at readthrough sites in some retroviral RNAs.

    Notes: Abstract A generally applicable system for targeted mutagenesis of a chromosomal sequence is described. The Escherichia coli tufA gene was mutated using a recombinant M13mp9 phage vector carrying a tuf gene. Integration via crossing over with the chromosomal tufA target gene produced an M13 lysogen. These lysogens were screened for resistance to kirromycin. The M13 phage carrying tufA mutations were efficiently retrieved by a genetic procedure. Genetic mapping was performed with the M13 vectors. The same recombinant M13 phage was used for mutagenesis, lysogen formation, gene replacement, retrieval, mapping and sequencing of kirromycin mutants. Three different mutations yielding resistance to kirromycin were found: two of these have previously been found and characterised, while the third mutation, Gly316 → Asp, is a new mutant. We also report the identification of a fourth kirromycin-resistant mutant, Gln124 → Lys.

    Notes: Abstract The FIS protein (factor for inversion stimulation) is known to activate the transcription of rRNA and tRNA operons in Escherichia coli. In this report the growth behaviour of a fis bacterium was studied under steady-state conditions and in competition experiments with a wild-type bacterium in a chemostat. The growth rate of the fis bacterium was reduced as compared to that of an isogenic wild-type and this reduction increased linearly with increasing growth rate. A similar result was obtained in competition experiments in a chemostat, with the selection factor increasing linearly with the growth rate.
