    Notes: Summary We cloned and sequenced two actin promoters from Physarum, and constructed plasmids carrying these promoters upstream of a bacterial chloramphenicol acetyltransferase (cat) gene. We then tested the plasmids for their ability to express cat in Physarum amoebae. We present reliable methods for introducing plasmid DNA into Physarum amoebae by electroporation, and show that expression of the cat gene in amoebae occurs in the presence, but not the absence, of one or the other Physarum actin promoter.

    Notes: Abstract Evidence is presented for the existence of two strong murine teratocarcinoma transplantation antigens (Gt) on the cell line PCC3. It is shown that the loci governing expression of these antigens are linked to the H-2 complex. These loci have been further mapped with respect to the brachyury marker (T) and H-2: Gt-1 lies 5±2 crossover units proximal to H-2 and 12±2 crossover units distal to T, Gt-2 lies 21±4 crossover units distal to H-2. It is possible that these strong transplantation antigens provide an embryonic analogue to the adult major histocompatibility system.

    Notes: Abstract Evidence is presented for the existence of teratocarcinoma transplantation antigens (Gt) encoded within the H-2 complex and present also on adult tissues. It has not been possible to separate these Gt loci from H-2 by recombination, and Gt factors map to each end of the H-2 complex. Previous reports indicating separation of all Gt loci from H-2 are reinterpreted. One class of such apparent recombinants has been shown to result from the outgrowth of tumor variants in mice of resistant genotype.

    Notes: Abstract We describe a genetic analysis of l(17)-2Pas, an embryonic lethal mutation on murine chromosome 17. Males transmitted the l(17)-2 allele to only 38% of their offspring, whereas females transmitted this allele at 50%. Two-point crosses revealed tight linkage between l(17)-2 and brachyury (T), and deletion mapping placed l(17)-2 outside of the hairpin-tail deletion (T hp ). To map this mutation more precisely, we intercrossed hybrid mice that carry distinct alleles at many classical and DNA loci on chromosome 17 and obtained 172 animals recombinant in the T to H-2 region. Strong positive interference was observed over the 14 cM interval from T to H-2K. Thus, a single recombinant can be informative; one such recombinant places l(17)-2 distal of the molecular marker D17Leh66D. Robust genetic maps can be constructed with multilocus crosses that share anchor loci. DNA markers can be interpolated onto these maps retrospectively.

    Notes: Abstract The spontaneous allele quakingviable (qk v ) exerts effects on myelination and spermiogenesis. The defects generated by qk v were not separated in a multilocus mapping cross that provided a mapping resolution of 0.1 centiMorgans (cM). Furthermore, no distortions suggestive of a large chromosomal anomaly associated with qk v were apparent. One plausible interpretation is that the quaking locus contains more than one functional domain, either organized into overlapping genes or expressed by alternative splicing mechanisms. The cloning needed to analyze this locus will be enhanced by the very high resolution of the meiotic mapping cross reported here. The recombinational distances on this qk v map were compressed compared with those previously reported in a high-resolution map for qk 1–1, an embryonic lethal allele of quaking induced by ethylnitrosourea. Additional crosses confirmed prior reports that the sex and the genetic background of the heterozygous parent can affect recombinational distances. These joint effects on recombination are strong enough to account for the discrepancy between the two maps. This variability of two-factor map values leads to the preferred multilocus map-building protocol discussed in the accompanying paper.

    Notes: Summary A method of indirect mutant selection, analogous to the bacterial penicillin technique, has been developed for Physarum polycephalum. The antibiotic netropsin was found to be lethal to growing but not to nongrowing (cycloheximide inhibited) myxamoebae. In reconstruction experiments, a 200 fold enrichment of cycloheximide sensitive over cycloheximide resistant cells was obtained. This technique was used to isolate temperature sensitive mutants. The mutant frequency among survivors was increased at least 40 fold by netropsin selection.

    Notes: Summary Cycloheximide-resistant mutants of Physarum polycephalum were induced in the haploid myxamoebae by the combined action of UV1 and caffeine (Haugli and Dove, 1972) or by treatment with NMG2. Eight independent mutants segregated in a Mendelian fashion (Table 1). Crosses between 6 of the mutants revealed 2 loci, actA and actB, for cycloheximide resistance (Table 2). All mutants are expressed in the plasmodium and are recessive in heterozygotes (Fig. 1 and 2). One mutation, conferring resistance to high levels of cycloheximide, was studied in heterokaryons and found to be incompletely recessive. An in vitro peptide synthesizing system was constructed from ribosomes from Physarum and supernatant factors from Saccharomyces cerevisiae. Cycloheximide strongly inhibited the activity of ribosomes derived from either wild type or mutants at the actB locus. In contrast, ribosomes from mutants at the actA locus were resistant to cycloheximide. Thus, the actA locus operates through the ribosomes.

    Notes: Summary Hybrid plasmids containing the bacterial resistance-transfer factor pBR322 and the yeast leu2 +gene have been used to isolate DNA fragments of Physarum that are capable of initiating DNA replication in a yeast host. Five of forty hybrid plasmids containing Physarum sequences transform leu2 -yeast to Leu+ at high frequency. The resulting Leu+ transformants are characterized by phenotypic instability. Supercoiled plasmid molecules containing pBR322 sequences can be detected in the transformed yeast, indicating that the transforming DNA replicates autonomously. Plasmid DNA isolated from Leu+ yeast can transform leuB bacteria. The hybrid plasmid recovered from the Leu+ bacterial transformants is identical to the original plasmid, indicating structural integrity is maintained during passage through the yeast host. These hybrid plasmids containing Physarum sequences have the same characteristics as those containing autonomously replicating yeast chromosomal sequences. As the temporal sequence of DNA replication is particularly accessible to study in Physarum plasmodia, the functional significance of these segments should be amenable to study.
