    Notes: Abstract N-glycans of neuropsin (serine protease in the murine hippocampus) expressed in Trichoplusia ni cells were released from the glycopeptides by digestion with glycoamidase A (from sweet almond), and the reducing ends of the oligosaccharides were reductively aminated with 2-aminopyridine. The derivatized N-glycans were separated and structurally identified by a two dimensional high-performance liquid chromatography (HPLC) mapping technique on two kinds of HPLC columns. Fourteen different major N-glycan structures were identified, of which 6 were high-mannose type (9.1%), and the remaining 8 were paucimannosidic type. The presence of insect specific N-glycan structures containing both α1,3- and α1,6- di-fucosylated innermost N-acetylglucosamine residue (23.3%), as below, was also confirmed by 600 MHz 1H-NMR spectroscopy.

    Notes: Summary A13C NMR study is reported of the hinge region of an intact mouse monoclonal antibody with a molecular weight of 150 K. Cys, Ile, and Pro analogs of the antibody labeled with13C at the carbonyl carbon were prepared by growing hybridoma cells in the serum-free media. Resonance assignments have been performed as described previously [Kato, K., Matsunaga, C., Igarashi, T., Kim, H., Odaka, A., Shimada, I. and Arata, Y. (1991)Biochemistry,30, 270–278]. The spectral data obtained show that13C NMR can give detailed information about the structure of the hinge region of the intact antibody molecule. Prospects for the future role of13C NMR in the structural analyses of larger proteins are briefly discussed.

    Notes: Abstract In an attempt to synthesize hydroxyapatite (HAP)/polymer composites, HAP crystallization was investigated in solution in the presence of ionic synthetic polymers. The side groups of the polymers used include carboxylate, dihydrogen phosphate, sulfate, and primary amine. Spontaneous HAP precipitation and amorphous–crystalline transformation occurring in both the presence or absence of ionic polymers were studied by measuring the solution turbidity change and titrating the released protons, respectively. The rates of HAP nucleation and growth were determined from an induction period before onset of crystallization and the subsequent propagation of HAP crystallites. The added anionic and cationic polymers, were found to suppress the crystallization in a concentration-dependent manner. An exception was a concentrated poly(acrylic acid), which was precipitated by calcium ion binding to accelerate the nucleation and the growth of HAP crystallites. These results suggest that a molecular interaction is operative between the ionic polymer chains and the growing HAP crystallites. Infrared spectroscopy and X-ray diffraction analysis revealed that the HAP precipitated in the presence of ionic polymers incorporated the polymer chains in the HAP microcrystalline aggregates. Based on these findings, HAP/poly(acrylic acid) composites were prepared through in situ HAP crystallization in the presence of poly(acrylic acid). Tensile testing of the composites and electron microscopic observation of their fractured surfaces revealed that the composite prepared through this wet process was superior to that obtained by simple physical mixing with respect to the wet state mechanical properties and homogeneity in mixing.

    Notes: Abstract A systematic method for 13C labeling of the glycan of immunoglobulin G for NMR study has been developed. A mouse immunoglobulin of subclass IgG2b has been used for the experiment. On the basis of chemical shift and linewidth data, it has been concluded that (1) the mobility of the carbohydrate chain in IgG2b is comparable to that of the backbone polypeptide chain with the exception of the galactose residue at the nonreducing end of the Manα1–3 branch, which is extremely mobile and (2) agalactosylation does not induce any significant change in the mobility. The results obtained indicate that even in the agalactosyl form the glycans are buried in the protein. Biological significance of the NMR results obtained is also briefly discussed.

    Notes: Abstract In order to understand the role of the glycans in glycoproteins in solution, structural information obtained by NMR spectroscopy is obviously required. However, the assignment of the NMR signals from the glycans in larger glycoproteins is still difficult, mainly due to the lack of appropriate methods for the assignment of the resonances originating from the glycans. By using [U-13C6,2H7]glucose as a metabolic precursor, we have successfully prepared a glycoprotein whose glycan is uniformly labeled with 13C and partially with D at the sugar residues. The D to H exchange ratios at the C1-C6 positions of the sugar residues have been proven to provide useful information for the spectral assignments of the glycan in the glycoprotein. This is the first report on the residue-specific assignment of the anomeric resonances originating from a glycan attached to a glycoprotein by using the metabolic incorporation of hydrogen from the medium into a glycan labeled with [U-13C6,2H7]glucose.

Es ist dem Verfasser gelungen, dasselbe Reagenz auf synthetische Polymerisatsysteme anzuwenden; Osmiumtetroxid kann zur elektronenmikroskopischen Darstellung von ABS- sowie ähnlichen kautschukverstärkten Kunststoffen als ein ausgezeichnetes Fixierungs- und Kontrastierungsmittel dienen. Die zur Untersuchung von ABS-Kunststoffen ausgearbeiteten Mikrotechniken werden auseinandergesetzt und die gewonnenen Ergebnisse werden unter Berücksichtigung ihrer weiteren Anwendung diskutiert.

    Notes: A phosphate-containing polymer was covalently immobilized onto a polyethylene (PE) rod, a poly(ethylene terephthalate) (PET) thread, and a PET film by a surface graft polymerization technique, followed by immersion in calcium phosphate solution to deposit a thin hydroxyapatite (HA) layer on the modified polymer surfaces. The PE rod had a tapered shape, while the PET thread was fixed with clips after implantation, both to minimize the micromovement which may occur in bone. The PE rod was implanted through press-fitting in the femur of rat. Significant enhancement was observed for direct contact of the implant surface with a newly formed bone for both the grafted only and the further HA-deposited PE rods in comparison with the untreated PE at 4, 5, and 6 weeks after implantation. The rats implanted with the modified PET thread in the femur were sacrificed 3 and 6 weeks after implantation. Statistically significant differences were observed for the untreated versus the grafted plus HA-deposited rods at 6 weeks after implantation. To study the resorption of the deposited HA on the methacryloyloxyethylene phosphate-grafted surface, HA-deposited PET films were subcutaneously implanted in the back of rats. The deposited HA was rapidly resorbed within 3 weeks of implantation. These results suggest that the phosphate polymer chains grafted on the PE and PET surfaces effectively induced nucleation and growth of HA crystals. It seems likely that the thin HA layer additionally deposited in vitro onto the grafted PE and PET surfaces was resorbed rapidly and then promoted the growth of HA crystals in vivo. © 1997 John Wiley & Sons, Inc.     Notes: Tumor promotion activity of polyethylene (PE) was estimated in terms of the inhibitory potentials on the gap-junctional intercellular communication using the metabolic cooperation assay. The gap-junctional intercellular communication of test cells was inhibited on the PE film, but this inhibitory activity was markedly decreased when the surface of the PE film was immobilized with collagen. These results suggest that the in vivo tumor promotion activity of the untreated PE may be stronger than that of collagen-immobilized PE. On the other hand, surface modification with RGDS peptide, which is well knownn as the sequence of cell attachment domain in extracellular matrix proteins, did not reduce the promotion activity of PE film. In addition, neither modification with bovine serum albumin nor RGES peptide reduced the activity of PE film. These findings suggest that reduction of the inhibitory activity on gap-junctional intercellular communication of collagen immobilization is not simply due to improved cell adherence via the RGDS sequence but to some cell-cell recognition via collagen molecules essential for the gap-junctional intercellular communication. © 1997 John Wiley & Sons, Inc.
