    Notes: Abstract Heat shock proteins (HSP) are a family of proteins expressed in response to a wide range of biotic and abiotic stressors. They are thus also referred to as stress proteins. Their extraordinarily high degree of identity at the amino acid sequence level and the fact that this cellular stress response has been described in nearly all organisms studied, make this group of proteins unique. We provide a brief historical overview of HSP research, as a background to summarizing what is known about HSP expression in fish. The expression of HSPs in fish has been described in cell lines, primary cultures of various cells, and in the tissues of whole organisms. Collectively, the data show that the expression of HSPs are affected in a wide variety of fish cells and tissues, in response both to biological stressors such as infectious pathogens, as well as to abiotic stressors such as heat and cold shock, and environmental contaminants. HSP research in fish is in its early stages and many studies are describing the expression of proteins in response to various stressors. Several studies have contributed to our understanding of the molecular nature and the molecular biology of HSPs in fish. Recent studies have shown a relationship between HSP expression and the generalized stress response in fish, but further research is needed to clarify the complex relationships between stress hormones and the cellular HSP response. In general, the HSP response seems to be related to the sensing of the stressor and the subsequent cellular effects which may adapt the cells to cope with the stressors. Consequently, such data may be of central importance in understanding the significance of HSP expression to the whole organism. We conclude with sections on laboratory methods used in HSP research and on potential applications of this knowledge in biomonitoring.

    Notes: Summary NMR-spectroscopic investigations of substituted dithiocarbacinic acid derivatives show the existence of configurational isomers relative to the C=N double bond, but no tautomerism. We applied chemical shift differences in the1H-,13C- and15N-NMR-spectra, N - H and C - H coupling constants and NOE difference measurements in the solvents CDCl3,DMSO-d 6 and partly CD3OD in order to discriminate between the isomers and to determine the configurational composition. Compounds4 and5 exist asE-isomers in all solvents, compounds1, 2 and3 as a mixture ofE/Z-isomers. InDMSO-d 6 theE-form is the preferred isomer.

    Notes: Abstract Cystic fibrosis (CF) is caused by mutations in the gene that encodes the cystic fibrosis transmembrane conductance regulator, CFTR. Previously we demonstrated that the common ΔF508 mutation in the first nucleotide binding domain (NBD1) alters the ability of the domain to fold into a functional three-dimensional structure, providing a molecular explanation for the observation that the mutant CFTR is retained in the endoplasmic reticulum and does not traffic to the apical membrane of affected epithelial cells. Notably, when conditions are altered to promote folding of the mutant protein, it can assume a functional conformation. Correcting the folding defect may have therapeutic benefit for the treatment of cystic fibrosis. Here we summarize these results and discuss the implications in vitro folding studies have for understanding the pathobiology of CF.

    Notes: Abstract The fatal autosomal recessive disease cystic fibrosis (CF) is caused by mutations in the gene which encodes the cystic fibrosis transmembrane conductance regulator (CFTR). Many of these disease-causing mutations, including the deletion of F508 (ΔF508) which accounts for approximately 70% of the disease alleles, occur in one of the two consensus nucleotide binding sequences. Peptide studies have directly demonstrated that the N-terminal nucleotide binding sequences bind adenine nucleotides. Structurally, circular dichroism spectropolarimetry indicates that this region of CFTR assumes a β-stranded structure in solution. The ΔF508 mutation causes a diminution in the amount of β-stranded structure and a concomitant increase in the amount of random coil structure present, indicating that either the mutant peptide has a different native structure or that the conformational equilibrium is shifted toward a more disordered form. Furthermore, the mutant peptide is more sensitive to denaturation, indicating that ΔF508 is a stability, or protein-folding mutant. Here we review these results and discuss their implications for interpreting the behavior of ΔF508in situ and for the rational design of new CF drugs.

    Notes: Dimensional analysis is applied to one form of fire growth in a compartment to derive two indices for the classification of linings, an ignitability index and a rate of heat release index, that can be combined into a fire growth parameter, which here is made proportional to the time to flashover in the internationally standardized Room/Corner Test. The ignitability index is the inverse of the time to ignition. The rate of heat release index is obtained by integrating the rate of heat release in time, weighted in such a way that the values of rate of heat release immediately after ignition are of higher importance than those at later times. Both indices are determined from test results of the Cone Calorimeter.

    Notes: Abstract The terminal steps of oxidative phosphorylation include transport of phosphate and ADP into the mitochondrial matrix, synthesis of ATP in the matrix, and transport of the product ATP into the cytosol where it can be utilized to perform cellular work. Three nuclear genome encoded membrane proteins, namely, the phosphate carrier (PHC), the adenine nucleotide carrier (ANT), and the ATP synthase complex, consisting of at least 13 individual subunits, catalyze these reactions. The locations of the α and γ subunits of the mitochondrial ATP synthase complex and the mitochondrial phosphate carrier, PHC, on human chromosomes were determined using cloned rat liver cDNA as probes. Human homologues of the α subunit are on chromosomes 9 and 18, the γ subunit are on chromosomes 10 and 14, and the PHC was localized to chromosome 12.
