    Notes: Abstract. We have isolated and characterized a genomic fragment encompassing the first six exons and 2.6 kbp 5′ flanking sequence of the rat cystathionine β-synthase (CBS) gene. A previously unknown exon approximately 3 kbp upstream of exon 1 was identified. The transcription start site was mapped to approximately 3 kbp upstream from the translation start codon and contains a consensus cap signal. The putative promoter region contains three GC boxes, in both orientations, and no TATA box. We have also compared a 1171-bp-long DNA sequence of the 5′ end of the rat CBS gene with the homologous mouse region of 1125 bp. We found two homologous Sp1 sites in the mouse gene and an overall sequence conservation of 70% with 88–89% similarity in the 80-bp regions surrounding the intron 0 splice sites.

    Notes: A mutant of Bacillus megaterium relieved from catabolite repression has been used to clone ccpA from B. megaterium by complementation. ccpA is the first gene of a presumed operon, in which it is followed by the motA homologue ORF1 and the motB homologue ORF2. The mutation maps in the 3′-terminal region of ccpA, where an in-frame duplication of 84 nucleotides located between two 9 bp direct repeats leads to an insertion of 28 amino acids near the C-terminus of CcpA. An in-frame deletion of 501 bp in ccpA exhibits the same phenotype as the 84 bp duplication. Deletion of ORF1 and ORF2 does not yield an apparent phenotype. A single-copy ccpA::lacZ transcriptional fusion is constitutively expressed, independent of whether the growth medium triggers catabolite repression or not. The ccpA mutation leads to relief of catabolite repression exerted by glucose, fructose, mannitol, glucitol and glycerol, whereas only smaller effects were found with ribose, citrate and glutamate. The respective growth rates on these carbon sources are uniformly reduced to a generation time of about 90 min in the ccpA mutant. Catabolite repression of a plasmid-encoded xylA::ccpA fusion is less efficient than that of a xylA::lacZ fusion in the same vector. Furthermore, overproduction of CcpA decreases catabolite repression of a singlecopy xylA::lacZ fusion approximately twofold. Thus, overexpression of CcpA may be counterproductive for catabolite repression, supporting the hypothesis that CcpA by itself may not bind sufficiently strongly to the cis-active catabolite-responsive element to exert catabolite repression.

    Notes: Abstract Polymer coatings are applied in many kinds of chemical sensors. The interaction with organic vapours changes the physical properties of the coating material. For optical sensors, changes in the coating volume and the complex refractive index are most important. Spectroscopic ellipsometry has been applied for the first time to the in-situ characterization of thin poly(dimethylsiloxane) films in contact with tetrachloroethene, toluene and cyclohexane vapours. The differences in bulk refractive index between organic solvent and polymer are large for toluene and tetrachloroethene and both effects were studied separately. Cyclohexane has a bulk refractive index very close to the investigated poly(dimethylsiloxane) films. Therefore the calculation of the volume fraction of the analyte in the mixture phase with the polymer is subject to large errors for vapour concentrations below 5000 ppm.

    Notes: Abstract The vast majority of probes used in fluorescence in situ hybridization (FISH) contain repetitive DNA. This DNA is usually competed out of a hybridization reaction by the addition of an unlabeled blocking agent, Cot-1 DNA. We have successfully removed repetitive DNA from two complex FISH probe sets: a degenerate oligonucleotide-primed polymerase chain reaction (DOP-PCR) single human chromosome library and genomic DNA. The procedure involved hybridizing in solution a DOP-PCR-amplifiable probe set with a 50-fold excess of biotin-labeled Cot-1 DNA, and capturing the Cot-1 DNA-containing hybrids using streptavidin magnetic particles, followed by purification and reamplification of the unbound fraction. Probes were checked for depletion of repeats by hybridization to chromosomes without Cot-1 DNA. Results showed hybridization patterns comparable to those achieved with untreated probes hybridized with Cot-1 DNA.

    Notes: Abstract Homocysteine is an independent risk factor for arteriosclerotic disease. Deficiency of cystathionine β-synthase (CBS) is the major cause of inherited homocysteinemia. The CBS gene is 25–30 kbp long and encodes a subunit of 63 kDa. The active form of the enzyme is a homotetramer that contains one heme and one pyridoxal 5′-phosphate per each subunit. It can also bind 1 mol of S-adenosylmethionine per mol of subunit. To date, an analysis of 205 homocystinuric alleles has been performed and 64 mutations found. The best studied, relatively “homogenous” patient populations are those of Ireland, Holland, and Italy. While the overall frequency of the two most frequent mutations is 24% for I278T and 31% for G307S, the breakdown between the countries varies greatly. For instance, the B6-nonresponsive G307S mutation accounts for 〉 70% alleles in Ireland and B6-responsive I278T mutation on the continent approaches 45%. In conclusion, further research is needed to define the mutations in individual countries to facilitate screening and genotype/phenotype correlations. Future biochemical studies will likely elucidate the role of heme in the enzyme and the tertiary structure of CBS.
