    Abstract: The use of explosives has led to a widespread distribution of 2,4,6-trinitrotoluene (TNT) and its by- and degradation products in the soil on former production and testing sites. The investigation of those large contaminated sites is so far based on a few selected soil samples, due to high costs of conventional HPLC and GC analysis, although huge differences in concentrations can already be found in small areas and different collection depths. We introduce a novel high-throughput screening system for those areas, which combines a smartphone-based collection of GPS data and soil characteristics with a fast MALDI-TOF MS quantification of explosives in soil sample extracts and finally a heatmap visualization of the explosives’ spread in soil and an analysis of correlation between concentrations and soil characteristics. The analysis of a 400 m2 area presented an extensive contamination with TNT and lower concentrations of the degradation and by-products aminodinitrotoluenes (ADNT) and dinitrotoluenes (DNT) next to a former production facility for TNT. The contamination decreased in deeper soil levels and depended on the soil type. Pure humus samples showed significantly lower contaminations compared to sand and humus/sand mixtures, which is likely to be caused by an increased binding potential of the humic material. No correlation was found between the vegetation and the concentration of explosives. Since the results were obtained and visualized within several hours, the MALDI-TOF MS based comprehensive screening and heatmap analysis might be valuable for a fast and high-throughput characterization of contaminated areas.

    Abstract: Noncovalent water-soluble nanocomplexes of C60 fullerene (C60) with chemotherapeutic drugs (Doxorubicin (Dox), Cisplatin (Cis), and herbal alkaloid Berberine (Ber)) were created. Their anticancer action toward various tumor cells was studied in vitro, addressing specifically their biological synergy, compared with the action of these drugs in the non-immobilized form. Different theoretical and experimental (SEM and AFM microscopy, UV-Vis, DLS, NMR and SANS spectroscopy, ITC calorimetry) methods were applied for getting insight into the nature of the nanocomplexes with drug molecules, as well as into the physical forces enabling stabilization of these complexes. Physicochemical mechanisms were proposed for drug interaction with C60. An enhancement of the toxic action of the created water-soluble C60-drug nanocomplexes toward cancer cells, compared to the action of free drug, was found. Specifically, the C60-Dox nanocomplexes demonstrated ~3.5 higher cytotoxic potential in the leukemic cell lines (CCRF-CEM, Jurkat, THP1, and Molt-16) in comparison with free Dox in the nanometer range of concentrations. Besides, C60 doubled the intracellular level of the up-taken Dox, which also evidenced its function as a nanocarrier. The toxic effect of C60-Cis nanocomplex toward Lewis lung carcinoma (LLC) cells was shown to be higher with IC50 values 3.3 and 4.5 times at 48 h and 72 h, respectively, as compared to the IC50 of free drug. 12.5 μМ Cis had no effect on LLC cells’ viability. The C60-Cis nanocomplex in Cis-equivalent concentration substantially decreased the viability of tumor cells, impaired their shape and adhesion, inhibited migration, and induced their accumulation in the pro-apoptotic sub-G1 phase of cell cycle. An induction of apoptosis by the C60-Cis nanocomplex was confirmed by the activation of caspase 3/7 and externalization of phosphatidylserine on the outer membrane of LLC cells after their double staining with the Annexin V-FITC/PI. The complexation with C60 promoted intracellular uptake of the Ber. An increase in C60 concentration in the C60-Ber nanocomplexes was accompanied by the elevation of their antiproliferative potential toward CCRF-CEM cells in the order: free Ber ˂ 1:2 ˂ 1:1 ˂ 2:1. These findings suggest a universal potential of water-soluble pristine C60 as a unique nano-platform for the delivery of the chemotherapeutic drugs in cytotoxic effect of these drugs.

    Abstract: In this work, the fabrication and characterization of a simple, inexpensive, and effective microfluidic paper analytic device (µPAD) for monitoring DNA samples is reported. The glass microfiber-based chip has been fabricated by a new wax-based transfer-printing technique and an electrode printing process. It is capable of moving DNA effectively in a time-dependent fashion. The nucleic acid sample is not damaged by this process and is accumulated in front of the anode, but not directly on the electrode. Thus, further DNA processing is feasible. The system allows the DNA to be purified by separating it from other components in sample mixtures such as proteins. Furthermore, it is demonstrated that DNA can be moved through several layers of the glass fiber material. This proof of concept will provide the basis for the development of rapid test systems, e.g., for the detection of pathogens in water samples.

    Abstract: Microscopic examination of human blood samples is an excellent opportunity to assess general health status and diagnose diseases. Conventional blood tests are performed in medical laboratories by specialized professionals and are time and labor intensive. The development of a point-of-care system based on a mobile microscope and powerful algorithms would be beneficial for providing care directly at the patient's bedside. For this purpose human blood samples were visualized using a low-cost mobile microscope, an ocular camera and a smartphone. Training and optimisation of different deep learning methods for instance segmentation are used to detect and count the different blood cells. The accuracy of the results is assessed using quantitative and qualitative evaluation standards.

    Abstract: Industrie 4.0 Ansätze haben sich in der fertigenden Industrie vielerorts etabliert. Big Data, IIoT und Künstliche Intelligenz (KI) ermöglichen neue Geschäftsmodelle und wirken sich dadurch auch verändernd auf Produktionssysteme aus. Mit dem Begriff der Industrie 5.0 formuliert sich ein menschzentrierter Ansatz. Die Praxisrelevanz dieser und ähnlicher Innovationen hängt maßgeblich davon ab, wie derartige Einsatzmöglichkeiten neuer, digitaler Technologien in die bestehenden Unternehmensstrukturen Einzug halten. Hierbei kommt es also auf Transfer und Akzeptanz neuer Technologien an. Digitalisierung gestaltet sich somit als sozio-technisches Problem. Dieser Beitrag gibt Einblick in aktuell verfolgte Transferansätze für Digitalisierung im Mittelstand und leistet einen Teil zu Theoriebildung, als dass er auch einen Ausblick auf weitere Aktivitäten in dem Umfeld liefert.

    Abstract: Silver halide polycrystalline infrared fibers (PIR) have unique properties such as excellent transmittance in the spectral range from 3 to 17 µm, while also being highly flexible, non-toxic, and non-hygroscopic. They are used in industry and medicine for CO2-laser power delivery, flexible infrared imaging, and remote process spectroscopy. While PIR fibers possess a quite low attenuation (0.1-0.5 dB/m) in the 8-12 µm range, their total transmittance is limited by significant Fresnel reflections at the fiber end faces due to the high refractive index of silver halide (〉2.1). Functionalization of these surfaces with specially designed Anti-Reflective Microstructures (ARMs) enables a striking enhancement of fiber transmittance. In this work, direct imprinting (or embossing) of microstructures to fiber ends and their profiling with a microstructured knife was applied to fabricate such ARMs. The resulting two-dimensional Moth-eye microstructures and one-dimensional microgrooves at the PIR-fiber ends enable to an increase of fiber transmittance in a broadband range of (5-17 µm) as well as to reach up to 20% improvement for PIR-fiber laser cables used for power delivery of CO2-lasers at 10.6 µm.

    Abstract: Direct optical detection methods such as surface plasmon resonance imaging (SPRi) and photonic-integrated-circuits (PIC)-based biosensors provide a fast label-free detection of COVID-19 antibodies in real-time. Each technology, i.e., SPRi and PIC, has advantages and disadvantages in terms of throughput, miniaturization, multiplexing, system integration, and cost-effective mass production. However, both technologies share similarities in terms of sensing mechanism and both can be used as high-content diagnostics at or near to point of care, where the analyte is not just quantified but comprehensively characterized. This is significant because recent results suggest that not only the antibody concentration of the three isotypes IgM, IgG, and IgA but also the strength of binding (affinity) gives an indication of potential COVID-19 severity. COVID-19 patients with high titers of low affinity antibodies are associated with disease severity. In this perspective, we provide some insights into how SPR and PIC technologies can be effectively combined and complementarily used for a comprehensive COVID-19 severity monitoring. This opens a route toward an immediate therapy decision to provide patients a treatment in an early stage of the infection, which could drastically lowers the risk of a severe disease course.

    Abstract: The first atomic layer deposition process for a ternary oxide is reported, which contains a metal of the platinum group, the delafossite PtCoO2. The deposition with the precursors trimethyl-Pt-methylcyclopentadienyl, Co-bis(N-t-butyl-N′-ethylpropanimidamidate), and oxygen plasma results in a process with a nearly constant growth rate and stoichiometric composition over a wide temperature window from 100 to 320 °C. Annealing of the as-deposited amorphous films in an oxygen atmosphere in a temperature window from 700 to 800 °C leads to the formation of the delafossite phase. Very thin films show a pronounced preferred orientation with the Pt sheets being almost parallel to the substrate surface while arbitrary orientation is observed for thicker films. The conformal coating of narrow trenches highlights the potential of this atomic-layer-deposition process. Moreover, heterostructures with magnetic films are fabricated to demonstrate the potential of PtCoO2 for spintronic applications.

    Abstract: Internationale Flughäfen sind wichtige Infrastrukturkomponenten mit der Funktion, vielfältige internationale Austauschprozesse zu ermöglichen. Dabei spielen Face-to-Face Kontakte über große Entfernungen hinweg eine Rolle, die für den Austausch vor allem von implizitem Wissen von großer Bedeutung sind. Bei der Realisierung dieser Funktion stehen Flughäfen in Wechselbeziehungen mit zahlreichen regionalen Entwicklungsbedingungen. Sie nehmen diese in Anspruch und entfalten hierbei vielfältige direkte und indirekte Wirkung auf ihr Umfeld. Auf diese Weise entstehen funktionale Räume, die als Flughafenregionen bezeichnet werden. Das vorliegende Working Paper beschreibt den Status quo der Flughafenregion Berlin-Brandenburg. Dem liegt eine Analyse zugrunde, die von einem Regionenverständnis geleitet ist, das system- und akteursbezogene Perspektiven aufweist. Sie geht weiterhin von der gemeinsamen These verschiedener Ansätze aus der Wirtschaftsgeografie und Innovationsforschung aus, die besagen, dass die Bedeutung räumlicher Nähe der Wissenserzeugung und -anwendung sowie des regionalen, interorganisationalen Lernens für Innovationsprozesse vorteilhaft sind. Charakterisiert wird die Flughafenregion deshalb als Wissenschafts- und Innovationsstandort. Gefragt wird darüber hinaus nach Ansätzen für eine Spezialisierung. Im Ergebnis wird deutlich, dass die Region bereits über starke Potenziale in diesen Bereichen verfügt, gegenwärtig eine hohe Ansiedlungsdynamik aufweist, aber noch kein typisches wirtschaftliches Spezialisierungsmuster ausgeprägt hat. Die zukünftige Entwicklung ist diesbezüglich offen. Im weiteren Verlauf der Szenarioanalyse wird daher der Frage nachgegangen, ob sich Spezialisierungspfade entwickeln werden, welche das sein könnten und wie regionale Entwicklungsbedingungen und Akteurskonstellationen beschaffen sein müssten, um einen solchen Profilierungsprozess zu befördern.

    Abstract: Label-free direct-optical biosensors such as surface-plasmon resonance (SPR) spectroscopy has become a gold standard in biochemical analytics in centralized laboratories. Biosensors based on photonic integrated circuits (PIC) are based on the same physical sensing mechanism: evanescent field sensing. PIC-based biosensors can play an important role in healthcare, especially for point-of-care diagnostics, if challenges for a transfer from research laboratory to industrial applications can be overcome. Research is at this threshold, which presents a great opportunity for innovative on-site analyses in the health and environmental sectors. A deeper understanding of the innovative PIC technology is possible by comparing it with the well-established SPR spectroscopy. In this work, we shortly introduce both technologies and reveal similarities and differences. Further, we review some latest advances and compare both technologies in terms of surface functionalization and sensor performance.

    Abstract: Endophytic fungi that colonize the plant root live in an environment with relative high concentrations of different sugars. Analyses of genome sequences indicate that such endophytes can secrete carbohydrate-related enzymes to compete for these sugars with the surrounding plant cells. We hypothesized that typical plant sugars can be used as carbon source by root endophytes and that these sugars also serve as signals to induce the expression and secretion of glycolytic enzymes. The plant-growth-promoting endophytes Serendipita indica and Serendipita herbamans were selected to first determine which sugars promote their growth and biomass formation. Secondly, particular sugars were added to liquid cultures of the fungi to induce intracellular and extracellular enzymatic activities which were measured in mycelia and culture supernatants. The results showed that both fungi cannot feed on melibiose and lactose, but instead use glucose, fructose, sucrose, mannose, arabinose, galactose and xylose as carbohydrate sources. These sugars regulated the cytoplasmic activity of glycolytic enzymes and also their secretion. The levels of induction or repression depended on the type of sugars added to the cultures and differed between the two fungi. Since no conventional signal peptide could be detected in most of the genome sequences encoding the glycolytic enzymes, a non-conventional protein secretory pathway is assumed. The results of the study suggest that root endophytic fungi translocate glycolytic activities into the root, and this process is regulated by the availability of particular plant sugars.

    Abstract: The acoustic pressure waves of ultrasound (US) penetrate biological tissues deeper than light. Another important feature of US its potential to generate light emission within the excited medium termed sonoluminescence. This promoted the idea of its use as an alternative energy source for photosensitizer excitation. Pristine C60 fullerene (C60), an excellent photosensitizer, was explored in the frame of cancer sonodynamic therapy (SDT). For that purpose, we analyzed C60 effects on human cervix carcinoma HeLa cells in combination with a low intensity US treatment. The time-dependent accumulation of C60 in HeLa cells reached its maximum at 24 h (800 ± 66 ng / 106 cells). Half of extranuclear C60 localized within mitochondria. The efficiency of C60 nanostructure’s sonoexcitation with 1 MHz US was tested with cell viability assay. A significant proapoptotic sonotoxic effect was found for HeLa cells. C60’s ability to induce apoptosis of carcinoma cells after sonoexcitation with US provides a promising novel approach for cancer treatment.

    Abstract: We propose a new mixed integer programming based heuristic for computing new benchmark primal solutions for instances of the PESPlib. The PESPlib is a collection of instances for the Periodic Event Scheduling Problem (PESP), comprising periodic timetabling problems inspired by real-world railway timetabling settings, and attracting several international research teams during the last years. We describe two strategies to merge a set of good periodic timetables. These make use of the instance structure and minimum weight cycle bases, finally leading to restricted mixed integer programming formulations with tighter variable bounds. Implementing this timetable merging approach in a concurrent solver, we improve the objective values of the best known solutions for the smallest and largest PESPlib instances by 1.7 and 4.3 percent, respectively.

    Abstract: Renewable energy sources are more important today than ever before, as they represent a key factor in reducing greenhouse emissions and fossil fuel consumption. Avoiding excessive burdens on future generations has now become the declared challenge of this century and justifies the efforts we must undertake to reduce global warming. This can only be achieved by conceiving new technical approaches, exploiting alternative resources and by raising people's awareness of the sustainable use of their own livelihoods. However, this change in awareness is a process that needs to be driven forward, especially in the field of research and education, and is therefore the declared aim of the Joint German-West African Conference on Sustainable, Renewable Energy Systems – SusRES. This annual conference provides a platform for exchange between scientists, students and many other interested parties, with technological progress at the forefront of the event. The SusRES conference is known as an accelerator for an ever-growing international network, not limited to the circumstances of a single country or even continent, which strives to take on and meet the global challenges of the present and future. This year's guiding theme of the event -Decentralized systems as a key factor in clean energy production for locations with limited infrastructure- of the event is primarily based on the consideration of how the energy supply of the future is to be designed. For some years now, a paradigm shift towards decentralised energy supply has been observed, which opens up new possibilities especially in areas with weak infrastructural supply. This is not just about providing energy from renewable resources, but also about exploiting this energy we have today efficiently and sustainably, whereby intelligent and interconnected systems play an ever-increasing role. The reader is provided with exciting insights into these topics within this conference proceedings elaborated by stakeholders from twelve time zones and four continents. In their contributions, the authors address important findings, especially from research but also from teaching, which represent an important milestone in their activities and thus make a significant contribution to our declared goals.

    Abstract: Processing biological materials with 3-dimensional (3D) printers has attracted increasing interest in several research areas. At the same time, off-the-shelf solutions are increasingly available in a wider variety to meet the current market demand. However, most of the available bioprinters are closed source, thus, modifications are quite challenging or require an avoidable consultation process with the manufacturer. Furthermore, the entry prices for basic machines amount to several thousands of euros. Whereas, high-end 3D bio printers with a vast array of features are available for several hundred thousand euros. Due to the immense potential of this tool in the field of biotechnology it is important to extend the availability of this technology for research purposes in terms of adaptability and price. This ongoing work focuses on open-source 3D printer heads with the ability to extrude biological materials. The print heads include several techniques to process low as well as high viscose biomaterials such as agar and gelatin. Additionally, obstacles such as continuous substrate tempering or integration into existing 3D printers are addressed. The work presented is open source and thus freely adaptable to any user’s specific needs. Our goal is to process a diverse range of biomaterials with different print techniques.

    Abstract: Optical-resolution photoacoustic microscopy (OR-PAM) combines high spatial resolution and strong absorption-based contrast in tissue, which has enabled structural and spectroscopic imaging of endogenous chromophores, primarily hemoglobin. Conventional piezoelectric ultrasound transducers are typically placed far away from the photoacoustic source due to their opacity, which reduces acoustic sensitivity. Optical ultrasound sensors are an alternative as their transparency allows them to be positioned close to the sample with minimal source-detector distances. In this work, a backward-mode OR-PAM system based on a planar Fabry-Pérot ultrasound sensor and coaxially aligned excitation and interrogation beams was developed. Two 3D imaging modes, using raster-scanning for enhanced image quality and continuous-scanning for fast imaging, were implemented and tested on a leaf skeleton phantom. In fast imaging mode, a scan-rate of 100,000 A-lines/s was achieved. 3D images of a zebrafish embryo were acquired in vivo in raster-scanning mode. The transparency of the FP sensor in the visible and near-infrared wavelength region makes it suitable for combined functional and molecular imaging applications using OR-PAM and multi-photon fluorescence microscopy.

    Abstract: The main aim of FE model updating is to iteratively correct the inaccurate parameters in a structural model so that the agreement between numerical predictions and modal test results is improved. Modeling the boundary conditions and contact interfaces between the different components is a challenge that needs to be taken into account in regards to the computational cost of the model for the iterative stage of the study. For instance, an extremely detailed model is of no use if each iteration takes hours to solve. A bottom-up approach can be followed to generate a highly accurate FE model by gradually increasing the complexity of the studied structure. This requires of a careful test planning phase to acquire experimental data from the structure in different boundary conditions and assembly stages. An overhead crane runway beam FE model is updated in this study case. Three FE models of increasing complexity in their boundary conditions and contact interfaces are optimized and updated with their three corresponding modal experimental datasets. This bottom-up approach allows dividing a complex problem in smaller phases, which results in a smaller number of design variables present on each model updating cycle. This is essential for the successful application of optimization algorithms with a limited computational power. In a qualitative sense, it allows the analyst to have a deeper understanding of the model from its conception and to have a solid grasp of the different what-if scenarios.

    Abstract: The present review defines the state of the art of parabolic trough collectors and identifies the concepts in the framework of innovation’s requirements. These are the increase of operating temperatures and of the power plant’s overall efficiency, as well as the cost reductions of the solar field by eliminating components and the reduction of parasitic power consumption (Pitz-Paal et al., 2007). The study investigates 34 collector concepts and classifies them according to their main structural features like mechanisms or materials. The categories include conventional collectors in Category A, collectors with alternative structure and sheet reflectors in Category B, with non-metallic materials like sandwich composite structures and concrete in Category C, with enclosed aperture in Category D and with fixed focus in Category E. For the technical-economic comparison, two conventional collectors vs. 14 innovations are analysed, following a design engineering method. The definition of a metric table defines the technical comparison criteria for those collectors suitable for thermo-oils or molten salts as heat transfer fluid. Results show that collectors operating with molten salts, instead of thermo-oil, have a significant techno-economic potential for utility scale application, for instance, the Molten Salt Trough, the UltimateTrough and the SkyFuelDSP (dispatchable solar power) as forerunners of their respective categories. Results show also that the inflatable Heliovis collector operating with thermo-oil has a relevant economic potential.

    Abstract: Faults in industrial chiller systems can lead to higher energy consumption, increasing wear of system components and shorten equipment life. While they gradually cause anomalous system operating conditions, modern automatic fault detection models aim to detect them at low severity by using real-time sensor data. Many scientific contributions addressed this topic in the past and presented data-driven approaches to detect faulty system states. Although many promising results were presented to date, there is lack of suitable comparison studies that show the effectiveness of the proposed models by use of data stemming from different chiller systems. Therefore this study aims at detecting a suitable data-driven approach to detect faults reliable in different domains of industrial chillers. Thus, a unified procedure is developed, to train all algorithms in an identical way with same data-basis. Since most of the reviewed papers used only one dataset for training and testing, the selected approaches are trained and validated on two different datasets from real refrigeration systems. The data-driven approaches are evaluated based on their accuracy and true negative rate, from which the most suitable approach is derived as a conclusion.

    Abstract: Over the course of the last decade 3D printing has become a more established technology in terms of prototype development (rapid prototyping). The current effort is focused on transferring this knowhow into a product driven approach in order to manufacture even small batch sizes more economic. In terms of this work, this idea is adapted for the development of injection molds (rapid manufacturing). Hereby, a hardened polymer is used to create a forming cavity instead of tool-steel. In order to fulfil the mechanical process requirements of micro injection molding such as form stability under temperature and pressure this cavity is nevertheless integrated into a metal housing. A first set of experiments has been carried out using this develop mold to verify the capabilities of the developed prototype as well as molding process. Based on these first results, an optimization is carried out to improve the next iteration of this molding tool.

    Abstract: Autonomous systems (AS) that relay Internet traffic are not equally well connected. The failure of just a tiny portion of them can render multiple sites inaccessible and disconnect multiple service providers from the global network, while targeted attacks can severely impact Internet connectivity. Modeling Internet topology and measuring Internet connectivity can help determine Internet vulnerabilities and improve Internet performance. With this in mind, we have redesigned and implemented a framework called CORIA that enables the analysis of Internet connectivity risks using large network graphs. The requirements we set for the design include technological extensibility, combination of different data sets, intuitive and customizable user interface, rich visual representation of results, and performance efficiency.

    Abstract: The article considers the problem of evaluating the effect of promotions held in a company. This problem remains relevant in our time, as evidenced by various studies on this topic. The authors of the article provides an algorithm for evaluating the promotions effectiveness based on the sales analysis of time series. In this case, promotions are considered as interventions that cause deviations from the main trend of the time series, which can be included in the model as a separate fictitious regressor. The paper also considers mathematical models for representing the intervention and the main characteristics of interventions. To evaluate the systematic component of the time series, it is proposed to use the Prophet forecasting package, since it has a number of advantages that allow one to more accurately isolate the effect of the ongoing campaign. The evaluation algorithm was tested on historical data for a specific product of the product line of stores at gas stations. The results of evaluating the effectiveness of the evaluated product.

    Abstract: To achieve climate goals and contain further global warming, it is inevitable to reduce CO2 emissions especially in energy consumption. A way to do so is by integrating renewable energy sources (RES) into an energy system’s power generation. However, there is no standard procedure to decarbonise a locally restricted system. Therefore, the various local conditions have to be analysed and taken into consideration. The authors propose a model-based approach to decarbonise the energy system of the island Föhr, Germany. This includes various collected data sets on local conditions such as climate data and heat and power demand. The data is used to represent the island’s energy system and design a model-based solution in a simulation software. The authors identify potentials by comparing costs and revenues by addressing the deployment of different RES technologies. One finding is that heat generation causes 91 % of CO2 emissions making it the major producer. However, with the designed solution, emissions could be reduced to a third.

    Abstract: Background Berberine (Ber) is a herbal alkaloid with pharmacological activity in general and a high anticancer potency in particular. However, due to its low bioavailability, the difficulty in reaching a target and choosing the right dose, there is a need to improve approaches of Ber use in anticancer therapy. In this study, Ber, noncovalently bound to a carbon nanostructure C60 fullerene (C60) at various molar ratios of the components, was explored against Lewis lung carcinoma (LLC). Methods C60–Ber noncovalent nanocomplexes were synthesized in 1:2, 1:1 and 2:1 molar ratios. Ber release from the nanocomplexes was studied after prolonged incubation at different pH with the liquid chromatography–mass spectrometry analysis of free Ber content. Biological effects of the free and C60-complaxated Ber were studied in vitro towards LLC cells with phase-contrast and fluorescence microscopy, flow cytometry, MTT reduction, caspase activity and wound closure assays. The treatment with C60–Ber nanocomplex was evaluated in vivo with the LLC-tumored C57Bl mice. The mice body weight, tumor size, tumor weight and tumor weight index were assessed for four groups, treated with saline, 15 mg C60/kg, 7.5 mg Ber/kg or 2:1 C60-Ber nanocomplex (15 mg C60/kg, 7.5 mg Ber/kg). Results Ber release from C60–Ber nanocomplexes was promoted with medium acidification. LLC cells treatment with C60–Ber nanocomplexes was followed by enhanced Ber intracellular uptake as compared to free Ber. The cytotoxicity of the studied agents followed the order: free Ber 〈 1:2 〈 1:1 〈 2:1 C60–Ber nanocomplex. The potency of cytotoxic effect of 2:1 C60–Ber nanocomplex was confirmed by 21.3-fold decrease of IC50 value (0.8 ± 0.3 µM) compared to IC50 for free Ber (17 ± 2 µM). C60–Ber nanocomplexes induced caspase 3/7 activation and suppressed the migration activity of LLC cells. The therapeutic potency of 2:1 C60–Ber nanocomplex was confirmed in a mouse model of LLC. The tumor growth in the group treated with 2:1 C60–Ber nanocomplex is suppressed by approximately 50% at the end of experiment, while in the tumor-bearing group treated with free Ber no therapeutic effect was detected. Conclusions This study indicates that complexation of natural alkaloid Ber with C60 may be a novel therapeutic strategy against lung carcinoma.

    Abstract: External stimuli are potent tools that Nature uses to control protein function and activity. For instance, during viral entry and exit, pH variations are known to trigger large protein conformational changes. In Nature, also the electron transfer (ET) properties of ET proteins are influenced by pH‐induced conformational changes. In this work, a pH‐controlled, reversible 310‐helix to α‐helix conversion (from acidic to highly basic pH values and vice versa) of a peptide supramolecular system built on a gold surface is described. The effect of pH on the ability of the peptide SAM to generate a photocurrent was investigated, with particular focus on the effect of the pH‐induced conformational change on photocurrent efficiency. The films were characterized by electrochemical and spectroscopic techniques, and were found to be very stable over time, also in contact with a solution. They were also able to generate current under illumination, with an efficiency that is the highest recorded so far with biomolecular systems.

    Abstract: Artificial Intelligence (AI) is becoming increasingly important in many industries due to its diverse areas of application and potential. In logistics in particular, increasing customer demands and the growth in shipment volumes are leading to difficulties in forecasting delivery times, especially for the last mile. This paper explores the potential of using AI to improve delivery forecasting. For this purpose, a structured theoretical solution approach and a method for improving delivery forecasting using AI are presented. In doing so, the important phases of the Cross-Industry Standard Process for Data Mining (CRISP-DM) framework, a standard process for data mining, are adopted and discussed in detail to illustrate the complexity and importance of each task such as data preparation or evaluation. Subsequently, by embedding the described solution into an overall system architecture for information systems, ideas for the integration of the solution into the complexity of real information systems for logistics are given.

    Abstract: Next Generation Sequencing technologies significantly impact the field of Antimicrobial Resistance (AMR) detection and monitoring, with immediate uses in diagnosis and risk assessment. For this application and in general, considerable challenges remain in demonstrating sufficient trust to act upon the meaningful information produced from raw data, partly because of the reliance on bioinformatics pipelines, which can produce different results and therefore lead to different interpretations. With the constant evolution of the field, it is difficult to identify, harmonise and recommend specific methods for large-scale implementations over time. In this article, we propose to address this challenge through establishing a transparent, performance-based, evaluation approach to provide flexibility in the bioinformatics tools of choice, while demonstrating proficiency in meeting common performance standards. The approach is two-fold: first, a community-driven effort to establish and maintain “live” (dynamic) benchmarking platforms to provide relevant performance metrics, based on different use-cases, that would evolve together with the AMR field; second, agreed and defined datasets to allow the pipelines’ implementation, validation, and quality-control over time. Following previous discussions on the main challenges linked to this approach, we provide concrete recommendations and future steps, related to different aspects of the design of benchmarks, such as the selection and the characteristics of the datasets (quality, choice of pathogens and resistances, etc.), the evaluation criteria of the pipelines, and the way these resources should be deployed in the community.
